SURVEYOR Mutation Detection Kits provide a simple and robust method to detect mutations and polymorphisms in DNA. The key component of the kits is SURVEYOR Nuclease, a member of the CEL nuclease family of mismatch-specific nucleases derived from celery. SURVEYOR Nuclease recognizes all insertions, deletions and base substitutions, and cleaves the mismatch of any heteroduplex DNA. Combined with our proofreading Optimase® Polymerase, non-specific background is significantly reduced.

Benefits
- Detection of all types of base-substitution and insertion/deletion mismatches.
- Size of cleavage fragments provides information about the location of the mutation.
- Cleavage products number indicates whether there are 1 or 2 or 3 or more mutations.
- Mutations can be detected in longer PCR products. 3-kb fragments have been tested.
- Pooling capabilities. To improve the throughput, PCR products can be pooled, cleaved with SURVEYOR Nuclease, and analyzed.

- All components other than the thermostable DNA polymerase and the PCR products are provided.
PCR products for analysis with SURVEYOR Nuclease should be prepared with a high-fidelity thermostable DNA polymerase, such as Optimase® Polymerase, to minimize the incorporation of errors that would result in background mismatches.
