The WAVE System Model 4500HT-HS is designed with higher through-put labs in mind and labs requiring the most sensitive analysis possible for low-level mutation detection.  The WAVE System is one of the most sensitive and accurate technologies for sequence variant analysis and separation of nucleic acid fragments. The WAVE 4500HT-HS System is fully automated, from sample collection to results analysis, and is able to analyze samples twice as fast as the standard WAVE 4500 System. Navigator™ Software creates the fragment specific methods, controls the instrument and analyzes your results. Fragment separation is achieved by the patented DNASep® HT Cartridge technology.


The fragments are intercalated with fluorescence dye and pass through the system’s fluorescence detector.  The fluorescence level is recorded over time, and this information is processed by the Navigator Software suite. It records the results in the form of chromatograms that can be further analyzed and filed by Navigator.

WAVE HSD - High Sensitivity Detection Module

Any WAVE 4500 System may be upgraded to a WAVE HS System by adding the WAVE HSD. The WAVE HSD consists of:

  • WAVE FDX - Fluorescence Detector (see description above)
  • WAVE HSX - High Sensitivity Accessory

The WAVE HSD incorporates fluorescence detection and intercalation chemistries to provide sensitivity enhancements for the detection of ds- and ss- DNA and RNA without fluorescently labeled primers. The HSD significantly increases signal sensitivity, typically by orders of magnitude when compared with UV detection.

The WAVE HS System can be used for applications that require extra sensitivity, including the following:

  • Detection of somatic mutations - enhances the detection of mutant alleles at low concentrations in the presence of highly abundant wild type alleles, in excess of 1:100 ratio
  • Pooling of DNA samples - detects mutations in plant samples (barley) at a 1:48 ratio of mutant to wild type plants
  • Analysis for samples originiating from PCR with low template concentration - PCR samples containing DNA template with concentrations as low as 0.5 ng can be successfully analyzed.

This accessory module may be added to a current WAVE 4500 System or a system upgraded to high sensitivity at the time of purchase.
