Practical Example
Detection of single
nucleotide polymorphisms that distinguish two strains of
mice.
The table below provides information about
a few SNPs that distinguish
the mouse strains C57BL/6J and 129/Sv. Primer sets were chosen to generate the PCR
products listed under “PCR Products”.
SURVEYOR Nuclease digestion of the heteroduplexed PCR products was predicted to generate cleavage fragments of the sizes listed
under “Digestion Products”. The presence and sizes of these cleavage fragments was verified using standard gel
electrophoresis (S),
fluorescent capillary electrophoresis (F), and the WAVE HS System (W). Experimental results obtained for the first 6 examples (framed in yellow) are shown in
the panels below the
table.
