High-Sensitivity Detection of Point Mutations and Tandem Duplications in Oncology Research

WAVE HS System Technology

The low abundance of mutated alleles in heterogenous samples poses problems for discovering mutations in early disease stages. Transgenomic has developed and is developing assays for mutations in various genes that are based on a single PCR amplification and analysis with the WAVE® HS System. These assays require neither fluorescently labeled primers nor any post-PCR purification. Detection and identification of mutations are below 1% of the total alleles present when using the WAVE HS System.

The WAVE HS analytical platform has a module for post-column intercalation and a fluorescence detector. The staining solution contains an intercalating dye, which will fluoresce only when bound to double-stranded DNA upon excitation with a Xenon lamp at 492 nm wave length and emission of 526 nm.
